If electroporation was previously performed at VARI and clones were generated, customers are responsible for notifying the gene targeting facility as to which ES cell clones should be microinjected. ES cells shipped to VARI for microinjection are required to undergo pathogen testing and karyotype analysis prior to injection. Clear instructions regarding the cells’ growth and selection history must be outlined before shipment.
Pathogen testing is performed by IDEXX-RADIL. Customers can ship the cells to IDEXX-RADIL for testing and send the Vivarium and Transgenics Core the paperwork demonstrating that the cells are pathogen-free. Alternatively, the customer can send the cells to VARI, where they will be cultured and sent as aliquots to IDEXX-RADIL for testing. The customer will be billed for the cost of the testing.
Karyotype analysis, chromosome counting and trisomy 8 FISH screens are performed by VARI’s Cytogenetics group. Customers can request spectral karyotyping (SKY) of ES cell clones at additional expense.
C57BL/6 or C57BL/6 albino blastocysts are used as hosts for ES cell clones that have a euploid karyotype. Injected blastocysts will be transferred to the oviducts of pseudopregnant foster mothers. Chimeric mice will be shipped to the customer at approximately three weeks of age at the customer’s expense.
